CarbPROFILE gels

CarbPROFILE gels are monosaccharides-Sepharose affinity matrices used for purification of specific carbohydrate-binding proteins. The carbohydrates are attached through their non reducing hydroxyl group after pre-activation of sepharose matrix by divinylsulfone (DVS) (see scheme 1 below). The binding of lectins and carbohydrates binding proteins to carbohydrate affinity gel is non-covalent and reversible with high capacity. Lectins and carbohydrates binding proteins are both usually stable compounds which can be recovered by competitive elution (i.e. 0.2 to 0.5 M of monosaccharide) or by modulations of pH and/or ionic strength in high yield and purity.

Preparation and quality control

Home made carbohydrate affinity chromatography media (CarbPROFILE) prepared in our laboratory.

  • Useful for purification of specific carbohydrate-binding proteins (i.e. lectins, lectins-like, GAG-binding proteins from naturals or recombinant sources).
  • High yield and purity under mild conditions (i.e. by competitive elution with corresponding monosaccharide or by modulation of pH and/or inonic strength).
  • Each CarbPROFILE gel is carefully controled according to our specifications:
  • Gel capacity (binding of validated standard over 15 mg/mL of gel)
  • Elution yield (over 85 % of injected standard recovered)
  • CarbPROFILE gel are available in 5, 10 and 25 mL slurry. Higher quantity are available upon request.
